There are still many mysteries that surround the lyophilization process but none can be any more elusive to our understanding than that of the interstitial region of the matrix (see INSIGHTs Vol. 2 No. 1 and Vol. 2 No. 2). This very unique region is formed as a result of the freezing process and is influential in determining not only the parameters of the lyophilization process but can seriously impact the design and construction of the freeze-drying equipment. In this INSIGHT I will (a), describe the formation of this region of the matrix; (b), why it is so deceptive in its nature and (c), the impact that it can have for those wishing to lyophilize a given substance.
